It was my very first time to buy 8 litres of skimmed milk at a time. The cashier was in shock when she saw the amount of milk to be purchased and she even asked the purpose of buying them. XD
I answered her that it was for experiment purposes and hard to explain in detail but they act as a medium for bacteria to grow. She joked as if I am the creator of life for them :)
To maintain sterility, the whole process was carried out in a horizontal laminar flow cabinet with flame on. The materials and appratus used were autoclaved and undergone UV treatment to prevent cross contamination.
The fresh milk was autoclaved to keep sterile and the inoculum from MRS broth (after 24 hours growth) was transferred into the milk for incubation.
It was kinda struggle to move the 8 litres of milk from level 3 to level 2 in order to transfer them into the incubator. Thanks God, we still transfer them safely.
After transferring into the incubator, we played some soft BGM and left a love note on the bottle just to ensure they will grow up happily.
Well, it sounds stupid but we believe it will work!
